Skin disorders and peptidoglycans—Beta-blocking drugs and bacterial peptidoglycan promote Th1-mediated inflammatory reponses in skin (Brain Behav Immun 2008 Jan[1]): The authors of this paper note that many inflammatory diseases such as psoriasis, atopic dermatitis, lichen planus, and vitiligo have also been associated with emotional stress and local alterations of the sympathetic (adrenergic) nervous system. Using a unique experimental model, the authors show that peptidoglycan from Gram-positive bacteria—but not lipopolysacharide from Gram-negative bacteria—promoted a local Th1-type proinflammatory response. Based on the responses elicited by adrenergic receptor (AR)-blocking drugs, the authors conclude that the Th1 polarization is mediated by inhibition of both beta1- and beta2-AR. The local pro-inflammatory response includes enhanced local expression of IFN-gamma, IL-12 and IL-23 as well as of IFN-beta and CXCR3 ligands. The local inflammatory response resulted in an increase of antigen-positive plasmacytoid dendritic cells (pDCs) in the draining lymph node. In sum, these results suggest that “altered sympathetic nervous activity together with selected pattern recognition receptors activation might serve as initiation and/or persistence factors for numerous Th1-sustained inflammatory skin diseases.” Psoriasis is a known Th1-sustained inflammatory skin disease.
